用來鑒別與多肽功能、穩定性和構象密切相關的特定的氨基酸位點。丙氨酸（Ala）是最小的手性氨基酸，通過將Ala系統性地、依次地替換序列中每一個氨基酸，來分辨出某一特定氨基酸對整個蛋白生物活性的影響。
